Ожирение является серьезной проблемой здравоохранения в России, где его распространенность растет. Для борьбы с ожирением используют различные методы, включая изменение образа жизни и медикаментозную терапию. Одним из препаратов, применяемых для лечения ожирения, является семаглутид, который стимулирует выработку инсулина и снижает аппетит. Разработка воспроизведенного препарата, содержащего семаглутид, является актуальной задачей, решение которой позволит увеличить доступность препаратов семаглутида в РФ.</p></sec><sec><title>Цель</title><p>Цель. Изучить сравнительную фармакокинетику, биоэквивалентность, безопасность и переносимость содержащих семаглутид препаратов GP40331 и Вегови в концентрации 0,68 и 3,2 мг/мл у здоровых добровольцев натощак.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Исследования проводили в параллельных группах здоровых добровольцев мужского пола с однократным введением исследуемых препаратов в дозе 0,25 мг (для концентрации 0,68 мг/мл) и 0,5 мг (для концентрации 3,2 мг/мл). Определение концентраций семаглутида в биообразцах сыворотки крови человека осуществляли с помощью метода высокоэффективной жидкостной хроматографии с тандемным масс-спектрометрическим детектированием.</p></sec><sec><title>Результаты</title><p>Результаты. Было показано, что значения 90%-х доверительных интервалов для отношений геометрических средних основных параметров фармакокинетики семаглутида составляют 90,22–110,29 и 86,48–108,98% для AUC0-t и Cmax семаглутида в концентрации 0,68 мг/мл и 90,62–115,71 и 92,86–113,51% для AUC0-t и Cmax семаглутида в концентрации 3,2 мг/мл. Была доказана сопоставимая безопасность исследуемых препаратов семаглутида.</p></sec><sec><title>Заключение</title><p>Заключение. Таким образом, по результатам настоящих клинических исследований препараты GP40331 и Вегови в концентрациях 0,68 и 3,2 мг/мл могут быть признаны биоэквивалентными и одинаково безопасными.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. Obesity is a growing public health issue in Russia, increasing the risk of cardiovascular diseases, type 2 diabetes, and hypertension. Controlling obesity involves lifestyle changes and pharmacotherapy. Semaglutide, an effective obesity treatment, stimulates insulin production and reduces appetite. Developing a generic semaglutide preparation will improve its availability in Russia.</p></sec><sec><title>Aim</title><p>Aim. To study the comparative pharmacokinetics, bioequivalence, safety and tolerability of semaglutide products GP40331 and Wegovy using concentrations of 0.68 and 3.2 mg/ml in healthy volunteers under fasting conditions.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. Bioequivalence studies, conducted per Good Clinical Practice, were open-label, randomized, and involved healthy male volunteers. Subjects received semaglutide at single doses of 0.25 mg (0.68 mg/ml) and 0.5 mg (3.2 mg/ml) under fasting. Bioequivalence was determined by the 90% CI of the ratios of geometric mean values of the primary pharmacokinetic parameters (AUC0-t, Cmax). Semaglutide concentrations were measured using high-performance liquid chromatography with tandem mass spectrometry.</p></sec><sec><title>Results</title><p>Results. The 90% CI values for the ratios of geometric means of the primary PK parameters of semaglutide were 90.22–110.29 and 86.48–108.98% (0.68 mg/ml) and 90.62–115.71 and 92.86–113.51% (3.2 mg/ml). Comparable safety was proven for both concentrations.</p></sec><sec><title>Conclusion</title><p>Conclusion.
