Локализованная склеродермия – редкое аутоиммунное заболевание, при котором происходит уплотнение отдельных участков кожи вследствие локального накопления белков дермального межклеточного матрикса, главным образом коллагена. Матриксные металлопротеиназы (протеолитические ферменты) – основные участники катаболизма коллагена, отвечающие за поддержание постоянства белкового состава дермиса. Изменения в активности и нарушения в экспрессии матриксных металлопротеиназ могут стать причиной развития болезни.</p></sec><sec><title>Цель</title><p>Цель. Оценить изменения в экспрессии секретируемых матриксных металлопротеиназ (ММП-1, ММП-2, ММП-3 и ММП-9) в пораженной склеродермией коже с перспективой разработки таргетных препаратов, используя метод количественной полимеразной цепной реакции.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. В работе исследовали биоптаты больных склеродермией, которые были получены из пораженной и непораженной болезнью кожи. Для оценки изменений генной экспрессии использовали метод полимеразной цепной реакции.</p></sec><sec><title>Результаты</title><p>Результаты. При анализе экспериментальных данных установлено увеличение экспрессии ММП-2 (3,379 ± 1,177) и ММП-9 (4,471 ± 1,836), а также снижение экспрессии ММП-1 (0,169 ± 0,036) и ММП-3 (0,240 ± 0,086). Показана строгая отрицательная корреляция (r = –0,93) между уровнем экспрессии ММП-1 и площадью поражения кожи. При проведении компьютерного анализа установлена взаимосвязь между изменениями в экспрессии ММП и особенностями поражения кожи.</p></sec><sec><title>Выводы</title><p>Выводы. Результаты работы подтверждают возможность использования ММП в качестве биомаркеров для оценки степени тяжести склеродермии и перспективного объекта (молекулярной мишени) для терапевтического вмешательства.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. Localized scleroderma is a rare autoimmune disorder characterized by a thickening of the skin due to the overdeposition of collagens and other proteins in the dermal extracellular matrix. Matrix metalloproteinases (MMPs) is a family of proteolytic enzymes, playing a crucial role in the catabolism of collagens, maintaining the protein composition of the dermis. Alterations in MMPs activity and expression contribute to the development of the disease.</p></sec><sec><title>Aim</title><p>Aim. To investigate changes in the expression of secreted matrix metalloproteinases (MMP-1, MMP-2, MMP-3, and MMP-9) in scleroderma lesional skin using quantitative polymerase chain reaction to identify potential therapeutic targets.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. Biopsies of lesional and unaffected skin were obtained from patients with scleroderma. PCR was used to assess changes in the expression of MMPs.</p></sec><sec><title>Results</title><p>Results. The analysis of experimental data revealed significant changes in the expression of MMPs: the increased levels of MMP-2 (3.379 ± 1.177) and MMP-9 (4.471 ± 1.836), and the decreased levels of MMP-1 (0.169 ± 0.036) and MMP-3 (0.240 ± 0.086).</p><p>The correlation analysis revealed a strong negative correlation (r = –0.93) between the expression of MMP-1 and the area of scleroderma lesional skin. The computer analysis suggested that changes in the expression of MMPs may contribute to the development of skin lesions as a factor.</p></sec><sec><title>Conclusions</title><p>Conclusions.
